چکیده

Urine SpecImens Diluted to Achieve Bluret-Predicted ProteIn Concentrations of 4 to 0.2 gIL The Coomassie Brilliant Blue (CBB) dye-binding method for determining total protein in urine is widely used in clinical laboratories because it is sensitive, easily automated, and free from interference by many clinically relevant substances (1-6). However, it yields different results for the same concentrations of different proteins (2,5-10). In clinical studies of urinary protein consisting predominantly of albumin, response curves reportedly were nearly linear for protein concentrations up to 1 gIL (2-4), but linearity for clinical specimens with mixed nonalbuminous proteins has not been established. We report here the accuracy and linearity of the CBB method for determination of urinary protein in patients with glomerular or renal tubular disease, in comparison with results obtained with the biuret method.

Determining total protein content of urine: centrifugation results in precipitation of the protein-coomassie brilliant blue dye complex.

The Coomassie Brilliant Blue (CBB) protein dye-binding assay (1) has been favorably assessed for determining the total protein content of urine (2-4), although discrepancies have been noted in its response to different proteins (5) and sample dilution (6). It is commonly accepted that the proteinCBB dye complex is soluble under assay conditions, but our studies reveal loss of color yield after ...

Rapid fluorometric estriol determination in pregnancy urine.

curves of precipitation are shown in Figure 1. Ampicillin, carbenicillin, amoxycillin, procaine penicillin, cephalothin, salicylic acid, acetylsalicylic acid, and dexamethasone caused no interference. The presence of methicillin in urine may be differentiated from the turbidity attributable to protein by measuring the absorbance at different intervals during the reaction, such as 0.5 and 5.0 mm...

Is standardization more important than methodology for assay of total protein in cerebrospinal fluid?

Four manual micromethods for protein determination, two turbidimetric (trichloroacetic and sulfosalicylic acid-sodium sulfate) and two colorimetric (Lowry and Coomassie Brilliant Blue--sodium dodecyl sulfate, CBB-SDS) were used to compare the standard curves for total protein (0.30 to 3 g/L) produced with three reference materials: bovine serum albumin, human serum albumin, and diluted human se...

A paradox clarified: set-point values for cholesterol, direct and enzymatic.

Lowry sensitivity and biuret specificity. Clin. In my review on cholesterol (1), I questioned the set points of an SMA reference serum of Technicon Instruments Corporation because the direct value, said to be standardized by the Abell procedure (2), and the value for the en-zymatic procedure, which should compare favorably to the Abell procedure, differed from each other by more than 30%. What ...

Optimization of Single Cell Protein Production by Aspergillus niger Using Taguchi Approach

: World population has been continuously raised and therefore due to this fact new source of foods, particularly proteins, are in demand. In recent years, single cell protein has been considered as an accepted substitute for animal and plant proteins. In this study, single cell protein production was studied in a batch submerged culture using Aspergillus niger PTCC5012. Experimental design was ...
